Dr. Josef Voglmeir博士长期致力于糖生物学和生物化学研究领域,英国曼彻斯特大学博士,在糖和糖肽修饰糖基转移酶的表达和定性等研究中成果显著。现工作于奥地利维也纳农业大学,中科院微生物研究所访问学者。
Seminar Abstract::
Glycosyltransferases are responsible for the molecular makeup of the glycome, the definition of their specificity is a central task in glycomics. In addition, these enzymes are also useful tools themselves for the generation of carbohydrate partners for binding studies. There has been great interest in using microarrays to generate glycopeptides structures enzymatically. This would allow either to probe the specificity of the enzyme against a panel of immobilized potential substrates, or to expand the diversity of the presented glycopeptide structures.
The development of an array platform based on self assembled monolayers of alkanethiols on gold helped to facilitate the synthesis of several glycan structures on gold surfaces by enzymatic glycosylation. Reactions were monitored by MALDI-ToF mass spectrometry and were found to be quantitative for several substrates. This work showed both the use of substrate arrays to explore enzyme’s activity and specificity in a label-free manner using MALDI-ToF MS read-out, and the application of an enzymatic strategy to obtain arrays of otherwise synthetically challenging glycopeptides.
